SNP Function
Intron variant. Subjects with the A allele at locus rs10521432 have two additional transcription factor binding sites. The differences may alter transcription rates of the MAOB gene. This could consequently lead to higher MAOB protein levels, which would result in lower monoamine concentrations in the CNS. These various in silico observations require confirmation by in vitro MAOB gene-expression studies.
Gene Function
The MAOB gene provides instructions for making the MAOB enzyme, which is located in the outer membrane of the mitochondria. It plays an important role in the breakdown of neurotransmitters and proteins which alter the flow of blood through blood vessels in the central nervous system and peripheral tissues. Like MAOA, it degrades dopamine. Alzheimer's disease and Parkinson's disease are both associated with elevated levels of MAOB in the brain. The normal activity of MAOB creates reactive oxygen species, which directly damage cells. MAOB levels have been found to increase with age, suggesting a role in natural age related cognitive decline and the increased likelihood of developing neurological diseases later in life. More active polymorphisms of the MAOB gene have been linked to negative emotionality, and suspected as an underlying factor in depression. Activity of MAOB has also been shown to play a role in stress-induced cardiac damage. The use of MAOB inhibitors to slow natural human aging in otherwise healthy individuals has been proposed, but remains a highly controversial topic. Supplements such as catechin and epicatechin and garlic show some ability to inhibit excess MAOB production. MAOB activity can increase for up to 60 minutes after exercise.
